We've been going for years - in fact, I'm on the Cape right now! What are your travel dates (approximate time of year) and what are your wishes? Outdoors/beaches/shopping? I suggest you pick one - Cape or MV. Message me - I can help you tons.
How are you getting here? Flying into Boston and renting a car? Driving from elsewhere? That makes a difference.
Most everything is casual, FYI - very little fancy schmancy. But you can go from cheap to expensive in lodging and dining. Do you have a hotel picked out or is this for a work related conference? If so, what part of the Cape? Give me a few particulars in your SWH, or PM me with more details. Glad to help!
Check tripadvisor.com and yelp.com not sure how people plan a (good) vacation without them.
I have found the coolest places through these sites and the rankings are spot on most every time.

Where are you staying? Ptown is great, but it is an all day trip if you are staying mid-Cape or west. I highly recommend Corporation Beach, eat at the Dennis Public Market location there. You can get amazing seafood and watch the sun set right on the beach. Since it looks like you are from the Midwest, I would do a whale watch! We prefer Dolphin Fleet, but there are lots. We saw a Great White in addition to lots of whales! There are some sunset/booze cruise types, especially off Ptown, lots of fun!
I haven't been to Cape Cod recently enough to reco places to eat. Provincetown, of course.

Cape is nice. Martha's Vineyard crowded, expensive and way too elitist for me.

Newport, RI is awesome as well if you're considering going up there. All of those towns get much less busy the first few weeks in September and the cost differential is amazing!!
Love the Cape!! haven't been there in like forever...so I'm not the best source right now....Although, my son went cycling and camping on Martha's Vineyard for a weekend with his scout troop last year and had a blast

It's been YEARS! The Cape is super peaceful and laid back, MV is busier and expensive.
I can't offer much more than that since I was a kid the last time I went.
Tripadvisor is a great site, but I also like to ask here because some of the hidden stuff isn't on that site.
Between AAA and local chambers of commerce, I've never had any trouble finding interesting things to do and see no matter where I go.
